The principle relies on the well-established indophenine reaction: thiophene reacts with an isatin reagent in a concentrated sulfuric acid medium to produce a deep blue complex. The intensity of this color is measured spectrophotometrically at 590 nm, where the specific absorption maximum occurs, and is directly proportional to the thiophene concentration in the original sample.
The required apparatus includes a spectrophotometer or filter photometer capable of precise measurements at 590 nm, matched absorption cells, and borosilicate separatory funnels. The procedure begins by adding a precise volume of the benzene sample to a separatory funnel. An exact quantity of the isatin-sulfuric acid reagent is then introduced. The mixture is shaken vigorously for a controlled duration to ensure complete extraction of thiophene and full development of the colored complex. Following a settling period to allow complete phase separation, the lower acid layer containing the blue complex is carefully drained. The absorbance of this extract is measured against a reagent blank prepared identically using thiophene-free benzene to compensate for any color contribution from the reagents themselves.
A calibration curve is established by carrying standard thiophene solutions through the same procedure. The concentration of thiophene in the unknown sample is then interpolated from this curve. Strict adherence to timing is crucial as the color complex can fade upon prolonged exposure to light or atmospheric moisture.
